TGF-b signaling pathway inactivation and cell cycle deregulation in the development of gastric cancer: Role of the b-spectrin, ELF
نویسندگان
چکیده
We have shown that loss of ELF, a stem cell adaptor protein, disrupts TGF-b signaling through Smad3 and Smad4 localization. Notably elf +/ /smad4 mice develop gastric cancer presenting this as an important model for analyzing molecular event in gastric carcinogenesis. To gain further insight into the functional role of ELF in gastric cancer suppression, we carried out a detailed characterization of cell cycle events leading to gastric tumorigenesis. elf / cells and elf +/ /smad4 mice demonstrate a marked alteration of cell cycle regulators, such as Cdk4, K-Ras, and p21. Levels of Cdk4 increased compared to normal controls, suggesting loss of ELF results in functional abnormalities in cell cycle regulation. We further demonstrate that the elf / MEFs show a disruption of G1/S cell cycle transition and a significant reduction in senescence. Thus, in response to ELF deficiency, the abnormalities of G1/S checkpoint and senescence contribute their increment of susceptibility to malignant transformation. 2006 Published by Elsevier Inc.

Effect of Transforming Growth Factor-β (TGF-β) on proliferation of gastric epithelial cells in culture
Objective: Helicobacter pylori has a well-established role in the development of gastric cancer. In vitro studies reveal increased proliferation of the gastric mucosa in the presence of H. pylori infection. It has been also shown that production of some cytokines, such as interleukin-1 beta (IL-1b) is in...
متن کاملInactivation of ELF/TGF-beta signaling in human gastrointestinal cancer.
TGF-beta/Smads regulate a wide variety of biological responses through transcriptional regulation of target genes. ELF, a beta-spectrin, plays a key role in the transmission of TGF-beta-mediated transcriptional response through Smads. ELF was originally identified as a key protein involved in endodermal stem/progenitor cells committed to foregut lineage. Also, as a major dynamic adaptor and sca...
متن کاملThe Role of Cyclooxygenase-2 in Signaling Pathways Promoting Colorectal Cancer
Colorectal cancer is one of the most common cancers in the world. Various factors are involved in the development and progression of this disease. One of these agents is cyclooxygenase-2 (COX-2). COX-2 is a product of the PTGS2 gene and converts free arachidonic acid to prostaglandins. COX-2 is not naturally expressed in most normal cells. Noticeably, the increased expression of COX-2 has been ...

Introduction: The incidence rate of liver cancer is continuously increasing. Currently, gene therapy is applied to improve various medical issues such as cancer treatment approaches. Correspondingly, fibromodulin involves in many biological and physiological processes through interaction with growth factors and signaling pathway receptors. The aim of this study was to investigate the effects of...
متن کاملTransforming Growth Factor-B Suppresses Nonmetastatic Colon Cancer through Smad4 and Adaptor Protein ELF at an Early Stage of Tumorigenesis
Although transforming growth factor-B (TGF-B) is both a suppressor and promoter of tumorigenesis, its contribution to early tumor suppression and staging remains largely unknown. In search of the mechanism of early tumor suppression, we identified the adaptor protein ELF, a B-spectrin from stem/progenitor cells committed to foregut lineage. ELF activates and modulates Smad4 activation of TGF-B ...
